Processing:

PAS-F (Periodic Acid-Schiff with diastase digestion):

The PAS stain reacts with the polysaccharides in fungal cell walls, coloring them pink or magenta, making them easily visible under a microscope. The diastase digestion step is sometimes added to remove glycogen and enhance the specificity of the stain for fungi.

GMS (Grocott’s Methenamine Silver Stain):

The GMS stain is another special stain that targets fungi, utilizing silver nitrate that binds to the fungal cell walls, resulting in a black or dark brown coloration against a light background. This contrast makes it easier to detect and identify the presence of fungi within a sample.
